Effect of 1α,25-dihydroxyvitamin D3 supplementation on peripheral blood polynuclear cells in Japanese Black cattle stimulated with lipopolysaccharide

Περίληψη
To investigate the effects of vitamin D on peripheral blood polymorphonuclear cells (PMNs) in cattle. PMNs were isolated from ten Japanese Black cattle, respectively. Each PMNs were divided into four groups, and cultured with 1α, 25-dihydroxyvitamin D3 (vitamin D group) or without 1α, 25-dihydroxyvitamin D3 (control group) under unstimulated, lipopolysaccharide (LPS) stimulated conditions. After PMNs culture, the cell viability was calculated by comparing with the number of live cells at the start of culture, and the mRNA gene expression of antimicrobial peptides and nitric oxide synthase in PMNs were investigated. The viability of PMNs was significantly higher in the vitamin D group than that control group under LPS stimulated conditions (p < 0.05). The relative gene expression of lingual antimicrobial peptide and beta-defensin 3, which are antimicrobial peptides, were significantly higher in the vitamin D group than those control group under LPS stimulated conditions, respectively (p < 0.01, p < 0.05). The gene expression of nitric oxide synthase was significantly higher in the vitamin D group than those control group under LPS stimulated conditions (p < 0.01). These results suggested that 1α, 25-dihydroxyvitamin D3 supplementation on PMNs in cattle might upregulate the innate immunity.
