Effects of L-Glutathione and Alpha-Tocopherol Added to Extenders on Sperm Quality in Cryopreservation of Epididymal Rat Spermatozoa

Resumen

Dondurma-çözme işlemleri, oksidatif stres nedeniyle hasarlı hücreye yol açan, artan reaktif oksijen türleri (ROS) üretimiyle ilişkili bir stres kaynağı olarak kabul edilir. Glutatyon, tüm memeli dokularında bulunan önemli endojen antioksidanlardan biridir. Alfa-tokoferol, sperm hücrelerini ROS hasarından koruyan ve lipid peroksidasyonunu önleyen doğal bir enzimatik olmayan antioksidandır. Bu çalışmada, farklı dozlarda %8 laktoz + %23 yumurta sarısı + %10 tris solüsyonu sperm uzatıcısına eklenen L-glutatyon (2,5, 5 ve 10 mM) ve alfa-tokoferolün (0,1, 0,2 ve 0,5 mM) sıçan spermatozoa sıvı azotunun dondurulması üzerindeki antioksidan etkileri araştırılmıştır. Dondurulmuş payet 37°C'lik su banyosunda 30 saniye çözüldükten sonra örneklerin spermatolojik analizleri yapılmıştır. Dondurma-çözme sonrası kontrol grubu ile karşılaştırıldığında en yüksek motilite 0,1 mM alfa-tokoferol (%23,89±1,82) ve 2,5 mM L-glutatyon (%28,00±2,71) gruplarında elde edildi (p<0,01). Ölü spermatozoa ve anormal akrozom oranları açısından gruplar arasında anlamlı bir fark bulunmadı (p>0,01). Kontrol grubu ile karşılaştırıldığında en yüksek plazma membran bütünlüğü 0,1 mM alfa-tokoferol ve 2,5 mM L-glutatyon gruplarında bulundu (p<0,01). Spermatozoanın dayanıklılığı için en yüksek motilite değerleri 0,1 mM alfa-tokoferol ve 2,5 mM L-glutatyon gruplarında kontrol grubuna göre belirlendi (p<0,01). Apoptotik ve nekrotik hücre oranı kontrol grubuna kıyasla 0,1 mM alfa-tokoferol ve 2,5 mM L-glutatyon gruplarında en düşüktü (p<0,01). Sonuç olarak, spermatozoa ekstansörüne 0,1 mM alfa-tokoferol veya 2,5 mM L-glutatyon eklenmesinin sperm hareketliliğini, fonksiyonel plazma membran bütünlüğünü ve hareketlilik dayanıklılığını artırdığı ve dondurma-çözme sonrası apoptoz değerlendirmesini azalttığı ve dolayısıyla rat spermatozoasının dondurma ekstansöründe başarıyla kullanılabileceği belirlendi.
